二色补血草LbDREB基因在酵母中的抗逆性分析

摘 要:

利用酵母表达系统研究了二色补血草的DREB基因(LbDREB)对不同胁迫的抗性。将LbDREB构建到酵母表达载体 pYES2中, 转化到酿酒酵母INVSc1菌株中, 并以转空pYES2质粒的酵母INVSc1 (pYES2)作为对照, 通过比较两种酵母在不 同胁迫下的存活率来研究LbDREB基因对NaCl、KH2PO4、Na2CO3、NaHCO3、低温、干旱、CuSO4和CdCl2胁迫的抗性。 结果表明, LbDREB转化的酵母在各种胁迫下的存活率均明显高于转空pYES2的对照酵母, 说明LbDREB基因除了具有传统 认为的抗旱、耐盐、抗寒的作用外, 还具有抗KH2PO4、Na2CO3、NaHCO3、CuSO4和CdCl2等胁迫的能力。

Stress Tolerance Analysis of LbDREB Gene from Limonium bicolor Kuntze in Yeast

Abstract:

To investigate the stress tolerance of LbDREB, the recombinant plasmid pYES2-LbDREB was constructed by inserting the LbDREB gene into the yeast expression vector pYES2. The recombinant plasmid pYES2-LbDREB was transformed into yeast Saccharomyces cerevisiae INVSc1, and the INVSc1 strain transformed with empty pYES2 was used as the control. The recombinant yeast INVSc1 harboring LbDREB(pYES2-LbDREB) and the control INVSc1 harboring empty pYES2 were respectively treated with NaCl, KH2-PO4, Na2CO3, NaHCO3, frezzing, drought, CuSO4 and CdCl2, and their survial rates were compared. The results showed that the survival rates of the recombinant yeast INVScl (pYES2-LbDREB) were obviously higher than those of the control strain under these stress conditions. It indicated that beside the known tolerance to salt, drought and cold, the LbDREB was also tolerant to NaCl, KH2 PO4, Na2CO3, NaHCO3, frezzing, drought, CuSO4and CdCl2 stress.
